IIC Services and Facilities

The services provided by the IIC fall into two major categories: cellular phenotyping by flow cytometry and cell sorting. Analytic procedures utilize FACSCalibur and LSR II multiparameter instruments. Sorting utilizes high speed multiparameter instruments available (FACSDiva and Aria II).

Cellular Phenotyping by Flow Cytometry

The core can perform the following analyses for DRC affiliates:

Identification and quantification of cell populations

  • T cells and T cell subsets
  • B cells and B cell subsets
  • Characterization of cells in adipose tissue (e.g. Macrophages and T cells)
  • Islet beta cells and islet endothelial cells

 

Cell Sorting

The IIC offers isolation of cells from different tissues through cell sorting to provide enriched or purified cell populations for the study of cellular function. Both training and operational assistance for the IIC sorting studies are provided through the IIC.

  • T cells and T cell subsets
  • B cells and B cell subsets
  • Enriched islet beta cell populations
  • Islet endothelial cells

 

Consultation

IIC directors provide extensive guidance regarding project planning, reagent selection and experimental approach.

Facilities

Laboratory

Flow cytometry equipment is available for IIC use at the University of Washington (UW) at South Lake Union (SLU), Benaroya Research Institute (BRI), Fred Hutchinson Cancer Research Center (FHCRC), and VA Puget Sound Health Care System (VAPSHCS). The IIC’s laboratories are located on the first floor of the UW SLU as well as the BRI, FHCRC, and VAPSHCS.

Equipment

IIC facility at BRI

  • 3 BD FACScalibur analyzing flow cytometers (4 colors);
  • 1 BD FACSort (FACScan) analyzing flow cytometer (3 colors);
  • 1 BD FACS DiVa high speed cell sorter system with thee lasers (8 colors);
  • 1 BD Aria II high speed cell sorter order system with 4 lasers (15 colors);
  • 1 BD LSRII system with 3 lasers (12 colors);
  • 1 BD LSRII system with 5 lasers (17 colors).

 

BRI provides technical assistance for the oversight and maintenance of these machines.

IIC resources at SLU

  • 1 BD Aria II high speed cell sorter order system with 4 lasers (15 colors);
  • 2 BD FACCANTO II analyzing flow cytometers (6 and 8 colors).

 

The UW Institute for Stem Cell and Regenerative Medicine (ISCRM) provides technical assistance for the oversight and maintenance of these machines.

IIC facility at FHCRC

The instruments that are available at the FHCRC Flow Cytometry Shared Resource include:

  • 3 BD FACSCalibur flow cytometers with two lasers (11 colors);
  • 1 BD FACS Canto I flow cytometer with two lasers (6 colors);
  • 1 BD FACS Canto II flow cytometer with three lasers (8 colors);
  • 3 BD FACSAria II cell sorters, two with 3 lasers (13 colors), and one with five lasers (18 colors);
  • 1 BD Influx cell sorter with five lasers (15 colors);
  • 2 BD LSRII flow cytometers with 4 lasers (18 colors) and 5 lasers (18 colors), respectively;
  • 2 Miltenyi Biotech AutoMACS pro magnetic cell separators.

 

Staff of the FHCRC Flow Cytometry Shared Resource provides technical assistance for the oversight and maintenance of these machines.

IIC satellite facility at VAPSHCS

  • 1 BD FACScalibur analyzing flow cytometer (4 colors)

 

The VAPSHCS provides a technician to oversee and maintain this machine.
