Reading R10 and Study Questions K were assigned last week. However, you are NOT RESPONSIBLE for question K.16.


Study Questions L are assigned after tomorrow's class (Tuesday, Mar 2). However, you are NOT RESPONSIBLE for question L.6.


The LAST READING, R11, on a human mutation affecting hand development, is assigned after Wednesday's class (Mar 3).

 

 

February 19 READING R9 Correction

Reading R9, which is assigned reading on Monday, Feb. 22, has an error. On page 5, left column, near the bottom, the large type that says.....

"(Note that in Figure 4 at position -34 the arrow should point downward, T -> C.)"

.....should not be there. The Legend to Figure 4 is correct. The upward arrow at position -34 means that the mutation results in a small increase in promoter activity.

(1). Study Question D.1 ANSWER.  The final product of the ligation is not
typed correctly, it should be:

(2). Lecture Notes page 43, Left Panel.  In Book 1 the spacing of letters
for item C is a bit strange and might be misread.  The phrase reads:

C. Comparison  of  cDNA  and  Genomic  DNA

(Note: "genomic DNA" refers to total (nuclear) DNA isolated directly from
cells of the organism.  "Genomic clone" refers to a piece of cloned DNA
isolated from a library made from an organism's total DNA, i.e., from a
genomic library).


(3). Lecture Notes page 44, Left Panel.  The Conclusion at the bottom
should read:

Conclusion:  INTRON information is present in the nuclear transcript and is
spliced out to make the mRNA.


(4). Reading R3. Page 3, 1st column, end of first paragraph. The sentence should end as follows: 

". . . . . skipping of exon 44, where the donor site of 
intron 43 is now joined to the acceptor site of intron 
44." (NOT intron 45).

Chi Squared Test

The description of the Chi Squared satatistical test in Reading R2 is not clear and does not follow common usage. A handout will be provided at class tomorrow (Tuesday, 1/12) which gives the Chi Square description from the earlier edition of "Introduction to Genetic Analysis". The description in the handout of degrees of freedom (df) is simpler, is the more commonly used one, and is the one used in the Study Questions. The value for df is taken as the number of classes of data minus 1. For the example in the reading of testing for independent assortment the number of classes is four (AB, ab, Ab, aB) and df = 3.
