Thanks, I have heard that from people using dos4gw.exe in another context and had failed to mention that, if it doesn't work in the root directory (C:\), try it in the same directory as the simul386.exe program. Well, I think you mean a rare allele, not a rare locus. Yes, if selection against the allele is relaxed, it will increase in the population. But not very quickly. There have been some nice papers on this -- the most accessible is probably Jim Crow's paper: Crow, J. F. The quality of people: human evolutionary changes Bioscience 16: 863-867 1966 Basically, if the gene was in equilibrium between mutation and selection, each generation enough copies were removed from the population by selection to compensate for the mutation rate, u. So when selection stops totally, all that happens is that the gene frequency increases by u per generation. That will, in most cases, be a very small percentage increase in the gene frequency. Calculate a case or two and you will see. For a recessive lethal gene, if the gene frequency is, say 0.0001, with mutation rate 10 to the -8, then in the next generation after selection ceases, the new gene frequency is 0.00010001.
