Objectives on the DeltaVision-RT Microscope
The following objectives are on the DeltaVision-RT deconvolution microscope:
20X UPlan S-Apo N.A. 0.75 (w.d. = 0.6 mm)
40X UPlan S-Apo N.A. 0.90 (w.d. = 0.6 mm)
60X PLAPON N.A. 1.42 Oil Immersion (w.d. = 0.15 mm)
100X UPlan S-Apo N.A. 1.40 Oil Immersion (w.d. = 0.13 mm)40X U-Apo N.A. 1.15 water immersion lens (w.d. = 0.6 mm)
60X UPlan S-Apo N.A. 1.20 water immersion lens (w.d. = 0.28 mm)
A 10X UPlan S-Apo N.A. 0.40 is also available (w.d. = 3.1 mm) -- ask Greg to put it on if you want to use it.
Notes:
The 100X oil objectives has an iris adjusted with a collar on the barrel of the objective. The collar should be turned full clockwise.
The 40X dry objective has a cover slip correction collar. Cover slips vary in thickness with a # 1 1/2 coverslip being nominally .17 mm thick. Adjust the collar for the best image: while looking at the specimen by eye, adjust the collar in small increments, refocusing continuously with the microscope's fine focus. Go for the highest contrast, "sharpest" image -- a small punctate structure should become blurry in an identical fashion as you defocus both above and below the structure.
