The UV/VIS collimator needs to bet set to provide correct Z-axis registration between the 405 nm excitation and the visible laser excitation wavelengths (488, 543, 633 nm).

From the main Zeiss toolbar

Select maintain

 

Select Maintain

 

 

Select Pinhole

 

 

 

 

Select Pinhole

From the Pinhole and Collimator window

Select UV/VIS

 

Do not adjust pinhole settings.

 

 

 

 

 

 

Under Collimator

Select UV/VIS

 

 

 

 

 

 

In the Collimator window

Input collimator setting

Input the following value, according to the objective lens being used, in the Position [mm] window:

0.05 for the 10X

-0.25 for the 20X

-1.25 for the 40X oil

-2.40 for the 63X oil

-1.30 for the 63X water

 

 

Do not use the 405 nm laser in combination with the visible lasers with the 100X oil objective.

Close the Pinhole and Collimator window. Your selection will be maintained.
