You should have an idea of how fluorescence works (http://micro.magnet.fsu.edu/primer/techniques/fluorescence/fluorhome.html), what fluorophores you will be using, and the full excitation and emission spectra of the fluorophores (http://www.probes.com/, http://www.clontech.com/index.shtml, http://www.jacksonimmuno.com/).
