Research summary

Lab work in Carl Anthony Blau’s lab in the School of Medicine (Hematology), University of Washington

The projects I am highly involved with include:

  • Identifying of the off-target effects of Epo on tumor progression and providing an method for regulating red cell production by administrating a chemical inducer of dimerization(CID), which binds to the modified ligand-binding site of engineered fusion proteins contained within hematopoietic stem cells.
  • ES cell self renewal project---Studying the signal transduction pathways in human embryonic stem cells

Methods such as quantitative rtPCR, western blot, multi-color flow cytometry etc are extensively practiced. Basic molecular experiments including transformation, maxiprep, miniprep, extraction of genomic/plasmid DNA, RNA from different sources are also broadly carried out. Lentivirus production for gene therapy(transduction&transfection) is done nicely. Notably, I got full training in purifying CD34+ cells from the cord blood and dog bone marrow (also linage depletion for bone marrow) and have been in charge of the purifying work with satisfying results.

Different mouse work such as intraperitoneal  injection, tail vein injection, blood collection from orbital sinus in the secured mouse facility of UW has been successfully conducted after rigorous training and exams.

Comprehensive tissue culture work has been practiced including culturing tumor cells/human ES cells.

Cell growth/drug inhibition tests are done by MTS assay and regular staining.

Fundamental hematology work WBC, RBC, platelets, hematecrits count/cytospin/ smear staining (Wright‘s and Giemsa’s) is carefully done.

Besides, I have been taking an active part in weekly lab meetings and other relevant meetings such as Gene Therapy Club including presenting updated research achievements and cutting-edge breakthroughs in related fields. My selection of certain articles has been adopted in one of the latest grant in the lab. (The critically important issue was first presented by me in one of the lab meetings)
