Primers are chosen such that the 3' end (which the DNA polymerase extends) is pointed in the right direction. The image below shows the primers as being outside the region to be amplified. However, if the sequence of the region to be amplified is known, the primers can be within the region itself, as described in Week 4 Practice Problems.
