(kan) Venus [plasmid pBES2]

Plasmid pBES2 is built from pDH22 except YFP has been mutated to the Venus variant. Those mutations are described in "A variant of yellow fluorescent protein with fast and efficient maturation for cell-biological applications" by Nagai, Ibata, Park, Kubota, Mikoshiba and Miyawaki in Nat Biotechnol 2002;20:87-90. In addition we have included Q69M, a mutation briefly mentioned in the Nagai et al. paper as potentially beneficial, although not necessary for the enhanced maturation. Plasmid pBES2 is coupled with pSH47 to introduce the Venus YFP at the N-terminus of a protein as described in Prein et al. The forward and reverse primers are the same as in pDH22. (The pictorial map is of pDH22, but the sequence is pBES2.)
